“Expression of OPG/ RANKL in macrophages (THP1) stimulated with endocannabinoids”

Abstract

Objectives: Evalute the expression of RANKL and OPG in macrophages stimulated with different concentrations of Anandamide. Methods: Macrophages (THP-1) were subjected to concentrations: 0, 5, 10, 20, 50 and 100M of Anandamide, quantifying by qPCR at different times, the expression of RANKL and OPG. The results are shown as a function of the relative expression of these genes (2-ΔΔCT) using the 18S gene as endogenous control. Results: Overexpression of OPG is observed at low concentrations of Anandamide, decreasing its expression once it reaches its peak, however no significant expression of RANKL is observed at any concentration. Conclusions: Anandamide is able to induce a dose-dependent expression by macrophages, acting in an osteoprotective manner at low concentrations. These findings suggest that the endocannabinoid system could be of value as a therapeutic target for the prevention and treatment of bone diseases, like periodontitis or conditions that involve remodeling of the bone tissue (graft).
