The implication of direct interaction between transcription factors pparγ and krüppel like factor 6 (klf6) in biomedicine: a bioinformatic perspective

Abstract
The KLF family of transcription factors hasattracted great attention in recent times, due to theversatility of functions that these genes and proteinscan perform in organisms. The implication of thesetranscription factors in important events such ascell growth, differentiation, migration, inflammationand angiogenesis is known. Its implication in thedevelopment of pathologies such as cancer is alsoknown. The aim of this articl e is to show the directinteraction that exists between the transcriptionfactor PPAR γ and consensus sequences in KLF6and its implication in biomedicine, seen from abioinformatic perspective. For this, a list of co-expressed genes for KLF6 was generated and thepromoter sequence close to transcription site start(TSS) was extracted. New consensus sequenceswere found using the MEME program, and thesequences similar to the new motif found weresearched through Tomtom Motif ComparisonTool and JASPAR CORE to assign a possiblefunction to the new motif found. The existence ofPPARγ binding sites in the KLF6 promoter wasevaluated using the EPD Eukaryotic promoterdatabase. Two PPARγ binding sites were found inthe promoter sequence of KLF6, in the regions-1264, -1451 upstream from the transcription startsite. This article preliminarily tests the hypothesisthat there is a direct interaction between thetranscription factor PPARγ and consensus sequences in the KLF6 promoter. This is consistent with aprevious finding, in which colon cancer cells weretreated with rosiglitazone, a PPAR γ agonist,resulting in increased expression of KLF6.
