Pilot evaluation of a clomipramine hydrochloride formulation for treatment of dogs with severe anxiety in Chile

Abstract

A pilot evaluation of the anxiolytic effects and safety of a clomipramine hydrochloride was carried out in dogs diagnosed with severe anxiety in Chile. Five dogs were selected based on standardized behavioral criteria, underwent medical screenings, and received clomipramine in a gradually increasing dosage up to 2 mg/kg daily for six months. Behavioral changes were assessed using a validated behavioral questionnaire and video recordings analyses. Results showed a significant reduction (50%) in total anxiety scores by the end of the study (p = 0.001). Specific behavioral improvements included reduced vocalizations, attention-seeking, repetitive walking, destructiveness and altered resting behavior (p < 0.05), with notable progress observed from the second month of treatment. No significant changes were found in inappropriate elimination or escape behaviors. Hematological, biochemical, and cardiac parameters remained within normal ranges throughout the pilot evaluation, indicating the drug's safety. Findings appear to support the efficacy and safety of a clomipramine hydrochloride for managing severe anxiety in dogs Chile, although further research with larger sample sizes is recommended.
