Oral presentation of the work: Study of the influence of human renal adipose tissue on tumor progression in kidney cancer Echegaray MD, Romeo LR, Bruna FA, Carón RW, Pistone Creydt V Tumor stroma is one of the emerging topics of interest in cancer research. In the present work, we evaluated: 1)adiponectin, ADAMTS1 and perilipin expression in conditioned media (CMs) of human adipose tissue explants from normal (hRAN) and tumor (hRAT) kidney; 2) AdipoR1, ObR, adiponectin and perilipin expression in hRAN- and hRAT. We also evaluated changes in the expression of: 1) AdipoR1, ObR, CD44 and vimentin in tumor (786-O, ACHN, Caki-1) and non tumor (HK-2) renal epithelial cells; 2) pERK/ERK and pPI3K/PI3K changes in cell lines incubated by 2 and 24h with hRAN-, hRAT, or control-CMs. Human renal adipose tissues were obtained from patients with renal cell carcinoma (hRAT, n=10) and kidney donors (hRAN, n=11). The CMs of hRAN and hRAT were collected 24h post incubation. Protein expression was evaluated by IHQ and WB. The cell lines were treated with CMs by 24h. Statistical differences were evaluated by one-way ANOVA or t-test, with Tukey´s post hoc tests. We observed a lower perilipin expression in hRAT-CMs vs. hRAN-CMs; and we found that adiponectin, AdipoR1, and perilipin expression were significantly increased in hRAN vs. hRAT explants (p<0.01). Also, we observed a decrease in the expression of AdipoR1 in ACHN and Caki-1 and the increased expression of ObR and CD44, when these cells were incubated with hRAT-CMs compared to hRAN- and control-CMs (p<0.001). In addition, we observed an increase in the expression of vimentin in HK-2, when these cells were incubated with hRAT-CMs. Further, we found an increase in the expression of pERK and pPI3K in HK-2 and 786-O, when these cells were incubated with hRAT-CMs compared to hRAN- and control-CMs (p <0.001). In conclusion, changes observed in the adipose microenvironment could be favoring tumor progression and therefore, the tumor stroma should be taken into consideration when dealing with a malignancy.
